First of all, we demonstrated here that a novel yvcE (cwl0) gene encodes a vegetative cell wall hydrolase classified into the DL-endopeptidase family. The C-terminal truncated protein, C-Cwl0-6His, retained cell wall hydrolase activity, and the substrate bond specificity indicated that it is a DL-endopeptidase in vitro. CwlO was secreted in culture medium during the vegetative growth phase. The disruption of CwlO, however did not affect cell growth, morphology, and motility. Second we investigated that the LysM domain of CwlE specifically recognized naked peptidoglycan in cell wall.
