To clarify the pathophysiology of hypercholesterolemia in renal diseases, we performed immunohistochemistry and reverse transcription-polymerase chain reaction to detect MCP-1and the adhesion molecules vascular cell adhesion molecule (VCAM)-1and intercellular adhesion molecule (ICAM)-1in kidneys from low density lipoprotein receptor knockout (LDLRKO)mice fed lipid-rich chow (n=12).They were harvested at 5 to 10weeks. For the control study, kidneys from wild mice fed lipid-rich chow and LDLRKO mice fed normal chow were also harvested.Severely high levels of hypercholesterolemia were seen in the serum of the LDLRKO mice,while no serum cholesterol elevation was observed in the wild mice.MCP-1 was enhanced in the glomerular endothelial cells and proximal tubular cells in the kidneys of LDLRKO mice, and this enhancement appeared prior to the expression of adhesion molecules and interstitial cell infiltration.
